Growth and Degeneration of Mouse Blastocyst In Vitro


Abstract
Objective:
@EN Little is known about the fate of embryo which is not normally growing in vitro. In this study, we investigated the degeneration process of embryos besides the normal growth.
@ES Materials and methods:
@EN Swiss Webster albino mice of 6~8 weeks old female and of 10~12 weeks old male were used. Blastocysts were flushed from the uterine horns with CMRL 1066 media containing 1mM glutamine and 1mM sodium pyruvate plus 50 IU/ml penicillin and
50mg/ml
streptomycin. Embryo growth and degeneration were examined and recorded everyday under a dissecting microscope.
@ES Results:
@EN The rates of normal development to early egg cylinder(EEC),late egg cylinder(LEC), and early somite(ES) were significantly decreased(p<0.01) as the duration of culture day increased. The incidence of embryo devoid of egg cylinder
increased(p<0.01).
as the duration of culture day increased. The production rate of abnormal egg cylinder also increased(p<0.01) as the duration of culture day increased.
@ES Conclusion:
@EN The analysis of the normal growth and dysmorphogenesis of the embryo from blastocyst up to ES would provide a useful experimental model for studying physical and chemical effects on the developing embryo.
